How much weight/relevance do things like lot of fortune, chiron, lillith dark moon etc., acthally have? Do they matter in a reading, or best to ignore? Or only when the information is supported elsewhere?

If I remember correctly, things like Chiron were not very relevant. On ToZ however you can find the relevant things. The new updates will surely be more in-depth as well.

Fixed degrees and stars are always there. They are normally in latent state if not aspected. If aspected they come into activated state. We all have every degree of the Zodiac, all 360, but not in all our charts every degree gets into play. Only when planet is conjunct or in less extent opposite to them, then they awake.
